Protocol summary

Study aim
Determining and comparing the effect of synbiotic beverage powder supplementation (containing pasteurized Akkermansia muciniphila, green tea extract, and glucomannan konjac gum) on changes in anthropometric indices (weight, waist circumference, waist-to-hip circumference, and body mass index), body composition (fat percentage, fat mass, fat-free mass, etc.), metabolic indices (lipid profile and fasting blood sugar), appetite, and blood pressure (SBP-DBP) in the intervention and control groups.
Design
A triple-blind randomized clinical trial with two parallel groups (intervention and control) will be conducted on 72 overweight or obese participants.
Settings and conduct
Among the clients in the nutrition clinic, 72 overweight or obese individuals will be randomly assigned to the intervention or control group after reviewing the inclusion and exclusion criteria. The participants will be given either a supplement or a placebo. Outcomes will be assessed at the beginning, middle, and end of the study.
Participants/Inclusion and exclusion criteria
Adults between 18 and 45 years of age who are overweight or obese without a specific medical condition, with no history of taking medications or supplements that affect the study outcomes or medications that interfere with green tea consumption, without a routine intake of high amounts of caffeine or fiber, or routine intake of dairy products and probiotic supplements, without a history of bariatric surgery, meal skipping, or a calorie-restricted diet, without a known allergy to the supplement components, and in a breastfeeding, pregnancy, or menopause state.
Intervention groups
The intervention group will receive three sachets of synbiotic beverage powder daily before every meal for two months.
Main outcome variables
Anthropometric indices and body composition

Randomization (investigator's opinion)
Randomized
Randomization description
Using random allocation software, a list of random blocks of 6 blocks of 6 is designed and a number is assigned to each block. The order of the contents of each block ensures a balanced allocation of individuals in the study groups. With each patient's visit, one of the blocks is randomly selected and the patient is assigned to one of the groups according to the order of its contents.
Blinding (investigator's opinion)
Triple blinded
Blinding description
In this triple-blind study, participants, main researchers, physicians, observers, data collection officials, evaluators, as well as data analyzers are unaware that each person participating in the study group is in the main intervention group (supplementation of the synbiotic) or placebo. For blinding, symbiotic supplements and placebo will be similar in appearance, packaging and labeling. The process of packing and coding of the supplement and placebo will be carried out by the manufacturer (Parsi Lact company).
